What is RT-PCR?

This stands forReverse Transcription Polymerase Chain Reaction, one of three tests for SARS-CoV-2. This test is not unique to this virus and has been used since the 1980s. The test is capable of detecting viral genetic material to determine its presence and has a reliability of over 90%, which is why it was initially the screening method recommended by the World Health Organization (WHO).

How does RT-PCR work?

It all starts with the use of a cotton swab or other device to extract biological material from the subject’s throat or nose. The material is stored with a buffer that breaks the cell membranes, allowing the material to escape to the outside. It would be expected that, in a sample from an infected person, the genetic material of the virus would be expected to leave the cell.

The material is then fed into a specialised machine to carry out the examination. Here, strands of viral ribonucleic acid (RNA) are detected by a strand of nucleic acid, called aprimer , which binds to a specific region. These primers contain a fluorescent marker attached. Subsequently, a reverse transcriptase (a protein with a particular role) extends the primer, creating complementary DNA, and releases the fluorescent marker.
